Does anyone have an easy way to align images taken with the two primary dichroic mirrors in the W1? I’ve found that there isn’t a great way to ensure that the hardware alignment between the 405-488-561-640 and 445-515-594 dichroics is spot on. So, we have to do post-imaging alignment when we are looking at fixed samples of DAPI-CFP-YFP-Cy5 or similar. Do you have a milled target that you image with both primaries in place and then come up with a shift correction? Do you use the parked disk? Do you have users include something like TetraSpeck beads for fiducial marks?
