Resonant scanning, is it quantitative even with a pixel clock?

When would be applicable cases to use:

  1. galvo vs resonant? is the speed wort the loss of quantitative data?
  2. single point (laser scanning) vs muliti-point (spinning disk) confocal? Is “less photobleaching” worth decreased resolution?
  3. water vs silicon immersion, do you really gain that much resolution?
  4. unidirectional vs bidirectional? is speed worth the “jaggedness” artifact that could be generated?

Does deciding the wrong modality really drastically change downstream data analysis for most experiments and/or users of core facilities? Or is it just about being “fast” and generating “pretty pictures”?
