When would be applicable cases to use:
- galvo vs resonant? is the speed wort the loss of quantitative data?
- single point (laser scanning) vs muliti-point (spinning disk) confocal? Is “less photobleaching” worth decreased resolution?
- water vs silicon immersion, do you really gain that much resolution?
- unidirectional vs bidirectional? is speed worth the “jaggedness” artifact that could be generated?
Does deciding the wrong modality really drastically change downstream data analysis for most experiments and/or users of core facilities? Or is it just about being “fast” and generating “pretty pictures”?
