Resolution too low - advice for image aquisition needed

Hi all,

I am trying to segment fluorescent puncta. All is well on those which are far apart, but there are areas where these are very close. I am using the Zeiss Image.Z2 with Apotome.2 100x objective. Puncta should be around 200um.

As you can see in the image below, these are difficult to distinguish. I am by no means a microscopy expert, but I wonder if this is because the resolution is too low? I am wondering which system it may be best to try from here on.

Could you clarify a bit more about which system you’re using - is it the Zeiss Axio Imager? What camera are you using?

Also, did you mean 200 um or 200 nm?
