Does airyscan detector rearrangement affect intensity?

I have just small question.

I usually use the LSM980 Airyscan for imaging cells.

When I change plates to image a different one, the Airyscan detector starts rearranging its position, and I can see a change in brightness.

(rearrangement means - airy scan detector adjustment: fiber positon)

I wonder if this process affects the intensity values of the samples?

I want to compare the relative intensity values between different plates.

It will. You need to realign the array each time you change your sample.

The coverglass bottoms of multi-well plates can be horribly unreliable with respect to thickness - and unless you are using high precision #1.5 (170um) plates you will notice quite a bit of variability in the alignment. Changing to high precision plates will help.
